ABSTRACT

Objective@#To clarify the genotype of varicella-zoster virus (VZV) in Jilin province in 2017, and to discriminate between vaccine strain and wild-type strain.@*Methods@#Vesicle fluid and throat swab samples were collected from 10 individuals with suspected VZV in Jilin province from January to March of 2017. Real-time fluorescent quantitative PCR was used to detect viral nucleic acid. Specific regions of ORF22, ORF38 and ORF62 of VZV were amplified by PCR. Viral genotype was determined by five SNPs of ORF 22 and vaccine strain or wild-type strain was distinguished by four SNPs of ORF 38 and ORF 62. The results were analyzed with MEGA5 and BioEdit software, using the VZV reference strain sequences from GenBank.@*Results@#VZV-positive strains were detected in 10 samples, all belonged to Clade 2. There was a synonymous mutation (C→T) in position 38 048 of JL17-7 strain. The nucleotide homology of ORF22 showed that all 10 samples were on the same branch with the Clade 2 referenced strains. Compared with Clade 2 referenced strains, the homology of nucleotide and amino acid for all 10 samples were 99.5%-100% and 99.3%-100%, respectively. The four specific SNPs of ORF38 and ORF62 in 10 samples were A-T-T-T, which were consistent with wild-type strain.@*Conclusions@#This study reveals that the VZV strains circulating in Jilin province in 2017 were all wild-type strains belonging to Clade 2.

ABSTRACT

Objective@#To explore the differences among three methods of nucleic acid extraction and three kinds of real-time fluorescence quantitative PCR instrument.@*Methods@#Twenty-five respiratory virus nucleic acid and 25 enterovirus nucleic acid positive samples were with selected at random and nucleic acids were extracted by using three methods (method A, B, and C). The results among different methods were analyzed by randomized block design. 25 respiratory viral nucleic acid positive specimens and enterovirus nucleic acid positive samples were detected by using three kinds of real-time fluorescence quantitative PCR instrument (instrument A, B, and C). The results among different instruments were analyzed by randomized block design.@*Results@#There was a significant difference among three methods of nucleic acid extraction in results(χ2=42.9162, P<0.001), in which method A and C had not significant difference(Z=0.837, P=0.3816>0.05), while method A vs. B, B vs. C were significantly different(Z=7.025, P<0.001; Z=7.9, P<0.001). There was also a significant difference among three kinds of real-time fluorescence quantitative PCR instrument in results(χ2=23.773, P<0.001), in which instrument B and C had no significant difference(Z=0.75, P=0.4533>0.05), while instrument A vs. B, A vs. C were significantly different(Z=5.70, P<0.001; Z=6.45, P<0.001).@*Conclusions@#There is difference among different methods and instruments in the test results under the same condition, which call for options in practical work according to need.

ABSTRACT

OBJECTIVETo explore the genetic etiology for fetuses featuring intrauterine growth anomalies using array-based comparative genomic hybridization (aCGH).

METHODSForty-nine fetuses were enrolled in this study. Genomic DNA of the abortive tissues was analyzed with aCGH.

RESULTSFourteen (28.6%) samples were found with chromosomal aberrations, which included 8 chromosomal aneuploidies and 6 micro-aberrations (4 with known clinical pathogenecity and 2 with unknown clinical significance).

CONCLUSIONNumerical and structural chromosomal aberrations underlie a significant proportion of fetal growth anomalies. aCGH has provided an effective method for delineating their genetic cause.

ABSTRACT

Objective To evaluate the effects of an intervention programme of health education and life style modification on postmenopausal osteoporosis women. Methods A total of 120 postmenopausal osteoporosis women were enrolled in this one-year randomized controlled follow-up study and assigned to the intervention group ( Group A, n = 60) or the control group ( Group B, n = 60). Both groups were treated with alendronate sodium. In Group A, education program was performed once a season in the form of face-to-face consultation or group session. In Group B, no additional intervention was used. The primary outcome was patients' compliance in follow-up. The secondary outcomes were change in bone mineral density (BMD).BMD was measured by dual-X-ray absorptiometry (DXA) on lumbar spine and hip at baseline and 12 months after the intervention. Results After one-year intervention,51 subjects in Group A and 38 in Group B completed the follow-up. Groups A showed better compliance. BMD on lumbar spine and hip was significantly increased in both groups when compared with baseline. The changes of BMD on lumbar (0.042+0.067 vs 0.026±0.070,P=0. O29) or Words region (0.029 +0. 129 vs 0.023±0. 143,P=0. 041 ) showed statistical significance between the two groups. Conclusion For alendronate sodium treatment, health management ensures the effectiveness of the therapy and improves the compliance of the patients.

ABSTRACT

Evidence is accumulating that aberrant microRNA(miRNA)expression contributes to the development and metastasis of cancer. Emerging evidence demonstrates that miRNA plays an important role in drug resistance in ovarian cancer and that it may become a new therapeutic target. It is important to further elucidate the mechanisms of miRNA associated drug resistance in ovarian cancer.

ABSTRACT

Objective To evaluate the effectiveness of comprehensive lifestyle modification in the primary health care system on status of pre-chronic diseases in a conventional healthcare program. Methods Two hundred and twenty-nine adult volunteers in 5 conventional healthcare centers were divided into 4 groups:group of prehypertension(n=67),group of prediabetes(n=38),group of abnormity serum cholesterol(n=71)and group of osteopenia(n=53).A muhicomponent behavioral intervention including education, physical activity, dietary practice, cease smoke, calcium supplement and exposure to sunlight was conducted for 12 months. The main outcome of each group was blood pressure, blood glucose, serum cholesterol and bone mineral density. Results Over 12 months, compared with the status before intervention, participants in each group were statistically significantly improved, the mean net reduction in systolic BP/diastolic BP was 9.4 mm Hg/6.6 mm Hg(1 mm Hg=0.133 kPa,t=5.93/8.29.P<0.05)in group of prehypertension;the reductions of blood glucose, serum cholesterol were 0.9 mmol/L(t=3.69,P<0.05)and 0.5 mmol/L(t=4.09,P<0.05)in group of prediabetes and group of abnormity serum cholesterol, and the increase of bone mineral density on lumbar spine was 0.015 g/cm2(t=2.03,P<0.05)in group of osteopenia,Conclusions For chronic disease prevention, ater one year the comprehensive lifestyle intervention produced beneficial changes in diet, physical activity, and biochemical parameters. This type of intervention is a feasible option to prevent chronic disease and should be implemented in the primary health care system.

ABSTRACT

Three hundred and ninety-five administrative staffs were randomly selected and provided health management to evaluate its effects in risk factors control of chronic diseases. At 1 year, the rate of people health knowledge and attitude were increased from 14.2% to 39.2%, 7.1% to 25.1%, P≤0.01 ; risk behavior exposure, including incorrect dietary intake, sedentary habit, alcohol consumption or smoking, reduced to some extent (P<0.05).
